Advanced off-gas measurement using proton transfer reaction mass spectrometry to predict cell culture parameters

چکیده
Background Mass spectrometry is a well-known technology to detect O2 and CO2 in the off-gas of cell culture fermentations. In contrast to classical mass spectrometers, the proton transfer reaction mass spectrometer (PTR MS) enables the noninvasive analysis of a broad spectrum of volatile organic compounds (VOCs) in real time. The thereby applied soft ionization technology generates spectra of less fragmentation and facilitates their interpretation. This gave us the possibility to identify process relevant compounds in the bioreactor off-gas stream in addition to O2 and CO2. In our study the PTR-MS technology was applied for the first time to monitor volatile organic compounds (VOC) and to predict cell culture parameters in an industrial mammalian cell culture process.

Advanced bioprocess monitoring by implementation of Proton Transfer Reaction - Mass Spectrometry (PTR-MS) for measurement of volatile components in the bioreactor
To demonstrate the proper functionality of the inlet system, three identical E. coli fermentations were conducted. The reproducibility of these experiments is shown by comparison of a set of process variables (Table 1, Figure 2). Figure 1: Simplified schematic of the setup. Fermenter off-gas is sampled and analyzed continuously. The inlet box, containing the flowcontrollers, is thermally contro...

We have developed an equilibrator inlet-proton transfer reaction-mass spectrometry (EI-PTRMS) system for the high-resolution measurement of the concentrations of multiple volatile organic compounds (VOCs) dissolved in seawater. The equilibration of six VOCs (dimethyl sulfide (DMS), isoprene, propene, acetone, acetaldehyde, and methanol) between seawater samples and the carrier gas, and the resp...

Output characteristics of fuel cells are affected by a large number of parameters such as geometry, dimensions, construction materials and conditions of supplying fluids. In this paper a mathematical model followed by a two-dimensional numerical approach has been presented to study the fuel cell parametrically. Effect of oxygen concentration at gas diffusion layer entrance, temperature and ...
متن کاملIdentification of the sulfoxide functionality in protonated analytes via ion/molecule reactions in linear quadrupole ion trap mass spectrometry.
A mass spectrometric method utilizing gas-phase ion/molecule reactions of 2-methoxypropene (MOP) has been developed for the identification of the sulfoxide functionality in protonated analytes in a LQIT mass spectrometer. Protonated sulfoxide analytes react with MOP to yield an abundant addition product (corresponding to 37-99% of the product ions), which is accompanied by a much slower proton ...

Chemostat cultivation is a powerful tool for physiological studies of microorganisms. We report the construction and application of a set of eight parallel small-scale bioreactors with a working volume of 10 mL for continuous cultivation. Hungate tubes were used as culture vessels connected to multichannel-peristaltic pumps for feeding fresh media and removal of culture broth and off-gas. Water...
